Semax Peptide is a synthetic heptapeptide with the sequence H-Met-Glu-His-Phe-Pro-Gly-Pro-OH, corresponding to the ACTH(4–7) fragment extended with a C-terminal Pro-Gly-Pro tripeptide. The compound was developed at the Institute of Molecular Genetics in Moscow and has been registered in Russia as a prescription nasal drop (0.1% and 1%) for cognitive and neurological indications. Semax’s research profile centers on neurotrophic signaling. A single application (50 μg/kg body weight) in rats produces a maximal 1.4-fold increase in BDNF protein levels and a 1.6-fold increase in trkB tyrosine phosphorylation in the hippocampus, suggesting the peptide modulates cognitive function through the BDNF/trkB system. Additional research has shown that intranasal Semax increases expression of both BDNF and NGF genes in the rat hippocampus, with Bdnf gene expression also increasing in the brainstem and cerebellum.

A 2025 study in the British Journal of Pharmacology identified a novel mechanism: Semax targets the μ opioid receptor gene Oprm1 to promote deubiquitination and functional recovery after spinal cord injury in female mice, suggesting a role in ubiquitination-dependent pathways beyond neurotrophin signaling.

Handling and storage
