PT 141, also known as bremelanotide, is a synthetic cyclic heptapeptide analog of α-melanocyte-stimulating hormone (α-MSH). Its structure incorporates a lactam bridge between the side chains of Asp and Lys, producing the cyclic conformation: Ac-Nle-cyclo(Asp-His-D-Phe-Arg-Trp-Lys)-OH. This modification confers resistance to enzymatic degradation and enhances receptor binding affinity.
PT 141 acts as a non-selective agonist at melanocortin receptors, with primary activity at the MC3R and MC4R subtypes, which are expressed primarily in the central nervous system. It exhibits little selectivity across the melanocortin receptor family, with the exception of MC2R (the ACTH receptor). The compound was originally identified as an active metabolite of melanotan II and was subsequently developed as a research tool for studying melanocortin signaling pathways.
