Kisspeptin-10, also known as KP-10 or Metastin 45-54, is a synthetic amidated decapeptide with the sequence H-Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH₂. It corresponds to the conserved C-terminal active fragment shared by all kisspeptin isoforms (KP-54, KP-14, KP-13, and KP-10), all of which exhibit the same efficacy and affinity for their receptor. It is the isoform most commonly used in biomedical research.

The peptide is the endogenous agonist of KISS1R (formerly GPR54), a Gq/11-coupled receptor expressed on gonadotropin-releasing hormone (GnRH) neurons in the hypothalamus. Binding triggers phospholipase C activation, inositol triphosphate and diacylglycerol production, and intracellular calcium mobilization. This signaling cascade stimulates GnRH release into the hypophyseal portal circulation, which in turn drives luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion from the anterior pituitary.

Kisspeptin-10 is the shortest of the four kisspeptin isoforms and has the shortest half-life, degrading in under 4 minutes in circulation. This short half-life makes it a useful tool for acute endocrine provocation studies, where its effects can be turned on and off in a controlled setting.

Note: Kisspeptin-10 is supplied as the C-terminally amidated form. A non-amidated form (CAS 478507-53-8) with molecular weight 1318.5 g/mol is used for rat studies. The human sequence is amidated at the C-terminus (Phe-NH₂), while some species variants differ. Verify the exact form, molecular weight, and purity against the Certificate of Analysis supplied with your lot.

Handling and storage

Reconstitute under aseptic conditions using sterile water or an appropriate aqueous buffer. Kisspeptin-10 is soluble in water. Aliquot immediately after reconstitution to avoid repeated freeze-thaw cycles. Lyophilized powder is stable for up to 2 years at −20 °C under sealed, desiccated, light-protected conditions. Reconstituted solutions should be stored at 2–8 °C and used within 14–21 days. Do not freeze reconstituted solution.
